Simple Single-Buffer Workflow — 15 Minutes The Zymoclean protocol is designed for speed and simplicity. Add the specially formulated Agarose Dissolving Buffer (ADB) directly to the excised gel slice, dissolve at 37–55°C, transfer to the Zymo-Spin™ Column, wash twice, and elute. The entire procedure takes approximately 15 minutes from gel slice to purified DNA — no organic extractions, no ethanol precipitation, no lengthy incubations.

Highly Concentrated Elution in Minimal Volume The Zymo-Spin™ column design permits DNA elution directly into as little as 6 µL of low-salt DNA Elution Buffer or water — yielding highly concentrated, sequencing-ready DNA from a minimal eluate volume. This is particularly valuable for downstream applications such as ligation reactions and next-generation sequencing library preparation, where high DNA concentration in a small volume is essential.

High Recovery Across a Wide Size Range The Zymoclean kit recovers DNA fragments from 50 bp to 23 kb with high efficiency. For fragments between 50 bp and 10 kb, recovery is typically 70–90%. For larger fragments between 11 kb and 23 kb, recovery is 50–70%. Up to 5 µg of total DNA per column can be recovered per prep.

Capped vs. Uncapped — What's the Difference? The D4001 uses uncapped Zymo-Spin™ I columns, which many researchers prefer for faster sample processing.
