Combining the tried-and-true alkaline lysis protocol with Zymo-Spin™ IIN column technology, the ZR Plasmid Miniprep-Classic delivers transfection-grade plasmid DNA in approximately 15 minutes — in a low elution volume of just 30 µL. The D4016 is the 400-prep bulk format, offering the best per-prep value for high-throughput labs and core facilities. Color-Coded Buffers for Error-Free Processing The P1 (Red), P2 (Green), and P3 (Yellow) buffers are color-coded for instant visual confirmation of complete bacterial cell lysis and neutralization at every step — eliminating one of the most common causes of failed minipreps. When the lysate turns completely yellow after P3 addition, neutralization is complete and the prep is ready to proceed.

Zymo-Spin™ IIN Columns — Transfection-Grade DNA at 30 µL The innovative Zymo-Spin™ IIN column features a unique design with zero buffer retention and a low 30 µL elution volume — delivering highly concentrated, transfection-grade plasmid DNA ready for immediate use. The column design features zero buffer retention and low volume (30 µl) elution. Zymoresearch Endotoxin levels are held to < 50 EU/µg, making the eluted DNA suitable for transfection of most standard cell lines.

15 Minutes from Culture to Plasmid DNA The complete protocol — resuspension, lysis, neutralization, column binding, endotoxin wash, plasmid wash, and elution — is performed at room temperature and takes approximately 15 minutes from bacterial culture to ready-to-use plasmid DNA.

Endotoxin Removal Step Included An Endo-Wash Buffer step is included in the protocol to remove endotoxins and produce plasmid DNA suitable for transfection with most standard cell lines — no separate endotoxin removal kit required.

ZR Plasmid Miniprep-Classic vs. ZymoPURE Miniprep Both kits isolate plasmid DNA from E. coli, but with different performance characteristics. The ZymoPURE Miniprep kit utilizes a modified alkaline lysis system in conjunction with a novel binding system, resulting in higher yields, higher concentrations, and lower endotoxin levels when compared to the ZR Plasmid Miniprep-Classic.
