Samples are digested with Proteinase K in a buffer matched to the sample type, then DNA is bound and washed on a midi-scale Zymo-Spin column designed to prevent buffer retention. Genomic, mitochondrial and viral DNA larger than 50 kb is recovered RNA-free, removing the RNase A step and supporting accurate quantification for library preparation, qPCR and arrays.
