Nuclei are first isolated from cells and then digested while still intact, using either of two included enzymes, Atlantis dsDNase or Micrococcal Nuclease. Digestion produces very homogeneous core nucleosome populations, and the associated DNA is then purified on Zymo Research spin columns. Each kit provides reagents for 20 preparations.
