This real-time PCR kit measures the percentage of methylation at custom genomic regions using restriction enzymes instead, offering a direct, straightforward assay for validating candidate sites.

Each DNA sample is split into a Test Reaction, which is digested with methylation-sensitive restriction enzymes, and a Reference Reaction, which is left undigested. Both are amplified by real-time PCR. Because unmethylated sites are cut and fail to amplify, the Ct difference between the two reactions is used to calculate percent methylation at the target region.
