The DWK Kimble® KONTES® 885300-0002 is a complete 2 mL all-glass Dounce tissue grinder set — the gold standard for gentle cell and tissue homogenization in molecular biology, biochemistry, and cell biology research. Manufactured from borosilicate glass 3.3, this instrument is designed primarily for cellular work where the nucleus remains intact after homogenization, producing a uniform cell lysate with minimal disruption to subcellular organelles. The complete set includes the glass tube and two glass pestles (large and small clearance) — everything needed to begin homogenization immediately upon receipt.

How Dounce Homogenization Works Tissue grinders produce homogenates by a combination of shearing and compression actions. The tissue sample is progressively ground into smaller pieces at the rounded end of the pestle as the spinning pestle is lowered into the tube. As the pestle is forced lower into the tube, the sample is displaced and forced between the straight outside wall of the pestle and the inside wall of the tube, compressing the tissue cells until they rupture. When the tube is pulled away from the pestle, a slight vacuum is created that pulls the sample back past the compression area, resulting in an additional homogenization stroke. Analytics Shop The degree of homogenization is controlled by the clearance between the pestle and tube, the rotational speed of the pestle, and the number of compression strokes.

Two-Pestle System — Large and Small Clearance The 885300-0002 includes two pestles with different clearance tolerances, used sequentially to achieve a controlled, progressive homogenization:

• Large clearance pestle (0.0030–0.0050 inch radial clearance) — used first for initial sample reduction, breaking down larger tissue chunks and cell aggregates into a coarse homogenate
• Small clearance pestle (0.0005–0.0025 inch radial clearance) — used second to form the final homogenate, achieving finer cell disruption while leaving nuclei largely intact

This two-stage approach gives the researcher direct control over the extent of homogenization and is the reason Dounce grinding is preferred over mechanical or ultrasonic methods when nuclear integrity or subcellular fraction preservation is required.

Borosilicate Glass 3.3 — Chemically Inert and Autoclavable Manufactured from borosilicate glass 3.3 Sigma-Aldrich, the KONTES® Dounce tissue grinder is chemically resistant to most biological buffers, detergents, and reagents used in cell lysis protocols. All glass components are autoclavable for sterilization between uses. Replacement components (tube and individual pestles) are available separately and are completely interchangeable across all units of the same catalog size.
