Cells are first grown in Expansion Broth (EB), where recombinant protein production is almost completely repressed, allowing healthy biomass to accumulate. The expanded culture is then transferred into Overexpression Broth (OB) to trigger high-level expression. This replaces standard LB-based procedures and works for many proteins.
