The 25-052-CI provides 5x lower trypsin concentration than the standard 25-053-CI — delivering effective enzymatic cell detachment with significantly reduced risk of protease-mediated cell damage, receptor cleavage, and viability loss for cell types that require a gentler dissociation approach.
The choice between 0.05% trypsin (25-052-CI) and 0.25% trypsin (25-053-CI) is one of the most important quality-of-care decisions in mammalian cell culture, particularly when working with primary cells and sensitive cell lines. Standard 0.25% trypsin is highly effective at detaching the broadest range of adherent cell types rapidly — but its potency can be excessive for cell types that detach easily or that are particularly sensitive to trypsin-mediated protein cleavage. Cells exposed to excess trypsin activity suffer cleavage of cell surface receptors, adhesion molecules, and signaling proteins; reduced post-passage viability; impaired re-attachment after passaging; and compromised downstream functional assays. The 0.05% trypsin concentration of the 25-052-CI provides sufficient enzymatic activity to effectively detach sensitive cell types while dramatically reducing these risks — making it the preferred format for primary cell cultures, embryonic stem cells, neurons, endothelial cells, and other sensitive or easily-detached cell types.
Like the 25-053-CI, the 25-052-CI is formulated in HBSS without calcium, magnesium, and sodium bicarbonate — eliminating divalent cations that would inhibit trypsin activity — and is Porcine Parvovirus Tested on every lot. The phenol red indicator provides visual pH monitoring, and the characteristic yellow color that may appear during shipping on dry ice can be corrected by adjusting pH to 7.2–8.0 with NaOH. IMPORTANT NOTE — COLOR CHANGE DURING SHIPPING
The phenol red indicator in Trypsin-EDTA gives the solution a pink color at the correct pH range of 7.2–8.0. During shipping on dry ice, the solution may change from pink to yellow due to CO₂ exposure — a normal shipping artifact. Adjust pH to 7.2–8.0 with NaOH to restore trypsin activity and pink color before use.
