Trypsin is a serine protease that cleaves cell-surface adhesion proteins — disrupting the cell-to-cell and cell-to-substrate bonds that anchor adherent cells to tissue culture vessels. The efficacy of trypsin is significantly inhibited by divalent cations (Ca²⁺ and Mg²⁺) commonly present in cell culture media and the cell culture environment. EDTA addresses this inhibition directly — as a chelating agent, EDTA sequesters Ca²⁺ and Mg²⁺ ions from the cell surface and culture environment, dramatically enhancing the speed and completeness of trypsin-mediated cell detachment. The 25-053-CI formulation is therefore prepared in HBSS specifically without calcium, magnesium, and sodium bicarbonate — ensuring no competing divalent cations are present to inhibit trypsin activity.

IMPORTANT NOTE — COLOR CHANGE DURING SHIPPING
The phenol red indicator in Trypsin-EDTA gives the solution a pink color at the correct pH range of 7.2–8.0. During shipping on dry ice, the solution may change color from pink to yellow due to exposure to CO₂ released from the dry ice — this is a normal shipping artifact and does not indicate product degradation. Adjust pH to 7.2–8.0 with NaOH to restore trypsin activity and the characteristic pink color before use.
