The 21-031-CV is the calcium and magnesium-free formulation — the critical format for cell washing prior to trypsinization and EDTA-based cell detachment, where the presence of divalent cations (Ca²⁺ and Mg²⁺) would interfere with enzyme activity and inhibit complete cell detachment.
Dulbecco's Phosphate-Buffered Saline was originally formulated by Renato Dulbecco to maintain cell culture media in the physiological pH range of 7.0–7.6 without the CO₂ dependency of bicarbonate buffers. The phosphate buffer system of DPBS maintains physiological pH across a wide range of temperatures and atmospheric conditions — making it stable outside the CO₂ incubator for extended periods, unlike bicarbonate-buffered media. The 21-031-CV serves as an excellent irrigating, transporting, and diluting fluid while maintaining cell tonicity and viability for limited time periods outside the incubator. It can also be used as a diluent for reagents, antibodies, and other biological materials requiring an isotonic, physiological pH buffer.
The 21-031-CV features a uniquely long 36-month shelf life — the longest in the Mediatech media lineup — and an unusually wide storage temperature range of 2°C to 30°C, making it suitable for refrigerator or ambient temperature storage and eliminating the strict cold chain requirements of other cell culture media.
