The 10-092-CV is the HEPES-buffered version of the DMEM/F-12 50/50 Mix — providing the additional pH buffering capacity of 15 mM HEPES beyond the bicarbonate buffer system, making it the preferred format for applications where stable pH maintenance outside of a CO₂ incubator is required, or where additional buffering capacity is needed to maintain physiological pH during handling, transport, or extended incubations.
The DMEM/Ham's F-12 50/50 Mix formulation was originally developed to combine the nutritional richness of DMEM — a modification of Basal Medium Eagle (BME) containing four-fold concentrations of the amino acids and vitamins found in BME — with the unique nutrient profile of Ham's F-12, which provides a wider range of amino acids, vitamins, trace elements, and growth factors at lower concentrations optimized for growth of nutritionally demanding cell types under reduced serum conditions. The 50/50 blend delivers a broader combined nutrient profile than either medium alone, making it the go-to basal medium for epithelial cells, endothelial cells, granulosa cells, primary cell cultures, stem cells, and any cell type that benefits from the richer nutrient environment of the combined formulation. Serum-free growth may be possible with appropriate protein supplements.
