The 10-090-CV is the preferred choice for labs running standard mammalian cell culture in a CO₂ incubator where the bicarbonate buffer system provides adequate pH control — and where the additional HEPES buffer of the 10-092-CV is unnecessary or undesired. The absence of HEPES makes the 10-090-CV the more economical and commonly stocked DMEM/F-12 format for routine CO₂ incubator-based cell culture workflows.
The DMEM/Ham's F-12 50/50 Mix formulation was originally developed to combine the nutritional richness of DMEM with the unique broader nutrient profile of Ham's F-12 — which provides a wider range of amino acids, vitamins, trace elements, and growth factors at lower concentrations optimized for growth of nutritionally demanding cell types under reduced serum conditions. The 50/50 blend delivers a broader combined nutrient profile than either medium alone, making it the go-to basal medium for epithelial cells, endothelial cells, granulosa cells, primary cell cultures, and stem cells. The 10-090-CV was originally formulated for rat neuroblastoma cells and MDCK (Madin-Darby Canine Kidney) cells, and has since been extensively validated across a wide range of demanding cell types requiring a richer nutrient profile than DMEM alone provides. Serum-free growth may be possible with appropriate protein supplements.
