RPMI 1640 — named for the Roswell Park Memorial Institute where it was developed in 1966 by George Moore, Robert Gerner, and Anne Franklin — is a modification of McCoy's 5A medium containing an increased level of inositol, along with a unique combination of salts, amino acids, and vitamins specifically optimized for the growth of human leukocytes and lymphoid cells. The 10-040-CV formulation is produced with L-glutamine and is the most commonly stocked RPMI 1640 format for routine lymphoid and suspension cell culture.
Unlike DMEM and most other classical basal media designed primarily for adherent mammalian cells, RPMI 1640 was specifically formulated for suspension culture of lymphoid and hematopoietic cells — the cell types that drive immunology, oncology, hybridoma technology, and monoclonal antibody production research. The RPMI 1640 formulation is uniquely suited to these cell types because of its lower calcium concentration (which favors suspension growth over cell adhesion), its higher phosphate content, and its specific amino acid and vitamin profile optimized for rapidly dividing lymphoid cells.
