Unlike standard RNA isolation kits that are optimized for cell culture or tissue inputs, the ZR Urine RNA Isolation Kit addresses the specific challenges of urine as an RNA source: cells and biological sediment are present at low concentration in a large-volume, dilute, high-salt matrix — requiring a specialized concentration step before RNA extraction that conventional spin-column kits cannot accommodate.
The ZR Urine RNA Isolation Kit's innovative workflow uses a uniquely designed syringe filter to concentrate cells and biological sediment from up to 30 mL of urine in a single step — filtering the full sample volume through a 1.6 µm ZRC GF™ filter membrane to capture all cells and sediment on the filter surface. Cells are then lysed directly on the filter using the specially formulated Urine RNA Buffer — a guanidinium thiocyanate-based lysis reagent specifically optimized for the unique sample matrix of urine. The resulting lysate can be processed immediately or stored for later transport and processing, providing flexibility for clinical sample collection workflows where immediate processing is not always possible.
The Urine RNA Buffer is also ideal for direct isolation of RNA from urinary microvesicles — submicron extracellular vesicles shed into urine by cells throughout the urinary tract that carry RNA biomarkers of urological disease, kidney function, and systemic conditions. One-step RNA isolation via Zymo-Spin™ IC Column matrix adsorption completes the procedure in less than 5 minutes, yielding high-quality total RNA eluted in as little as ≥6 µL — the smallest elution volume available for a urine RNA kit — for maximum concentration and RT-qPCR sensitivity.
